Melanotan 2 is a synthetic analogue of the naturally occurring α-melanocyte-stimulating hormone that results in a safe increase in the amount of natural melanin in healthy adults.

Melanotan 2 acts as a non-selective agonist of the melanocortin receptors, MC1, MC3, MC4, MC5. It produces melanogenesis, the production of the pigment melanin, through the activation of the MC1 receptor.
Both Melanotan and Melanotan II have been shown in the clinical setting to increase pigmentation even without exposure to UV light. However, the process of tanning progresses much more quickly with UV exposure.
The increase in pigmentation is largely proportional to the concentration of melanocytes within certain areas of the skin and as such areas with higher concentrations of melanocytes will see greater increases in pigmentation.
The cyclic nature of this peptide means that it is not quickly broken down and degraded by natural proteases and even just a few relatively low dosages can be administered to achieve a significant effect.
Even patients given only five low doses of this peptide every second day saw significant tanning activity in a 1996 study. An additional reported benefit of this peptide has been an increase in libido and sex drive in adult subjects.
It was initially developed as a drug candidate for female sexual dysfunction and erectile dysfunction. These observed sexual effects are thought to be related to its ability to activate the MC4 receptor, though the MC3 activation may also play a role.
